Principal Scientist, Bioconjugation – Platform Development

Role Summary:

The Principal Scientist is a highly motivated member of the Platform Development team and has a background in chemistry, biochemistry, or a related field to design, synthesize, and characterize complex bioconjugate therapeutics as part of our Platform Development group. This individual has broad opportunities to contribute and lead, from expanding our platform to encompass new targeting and payload modalities, to supporting the discovery of new FORCE platform candidates, all the way to scaleup of candidates entering development. The person in this role has experience in bioconjugation of proteins and/or nucleic acids and purification and analytical characterization of biopolymer conjugates. They collaborate closely with additional chemistry, biology, and research teams, supporting all stages of preclinical therapeutic development. This role will use creative and proactive problem-solving strategies to progress the design and development of Dyne’s therapeutic pipeline.  They act as an effective technical expert and work in a collaborative, constructive and mission-oriented focus for improving the lives of patients.

This role is based in Waltham, Massachusetts, without the possibility of being a remote role. 

Primary Responsibilities Include:

  • Prepare conjugated proteins and nucleic acids using an array of bioconjugation chemistries, including lysine acylation with active esters, thiol-maleimide labeling, and bio-orthogonal chemistry
  • Utilize macromolecular purification techniques including preparative SEC and HIC
  • Characterize conjugates and intermediates using analytical assays including UV-vis, SDS-PAGE, RP-HPLC, LC-MS, ELISA, and SPR
  • Independently analyze results, troubleshoot experiments, and develop new synthetic and analytical methods and protocols
  • Collaborate with biologists and pharmacologists to interpret in vitro and in vivo assays results and propose to advance Dyne’s technology
  • Provide mentorship to junior scientists in the Platform Development group
  • Thoroughly document results and provide clear presentations to diverse audiences of scientific and business collaborators
  • Prepare manuscripts and other documents for publication and/or regulatory submission

Knowledge & Skill Requirements:

  • Ph.D. in Chemistry, Biochemistry, Chemical Engineering, or a related field, ideally with 5+ years of research experience; or M.S. with 9+ years; or B.S. with 12+ years
  • Hands-on experience with bioconjugation, including purification, and characterization
  • Experience with bioconjugate therapies or drug delivery modalities such as targeted oligonucleotides, targeted radioligands, or antibody-drug conjugates (ADCs)
  • Experience on scale-up manufacturing supporting early development research a plus
  • Ability to critically evaluate, summarize, and interpret data
  • Excellent written and verbal communication skills, ability to make effective and compelling presentations in both large and small groups and communicating complex scientific and nonscientific issues to program teams
  • Understand published scientific literature outside of their immediate area of expertise and apply this understanding to drive innovation
